Toxicity of L-asparaginase drugs in acute lymphoblastic leukemia treatment

چکیده
Currently no therapy of acute lymphoblastic leukemia is conceivable without L-asparaginase drugs, with its antileukemic effect by extracellular asparagine depletion, thus preventing admission to leukemic cell. Besides high antitumor effect, drugs have side and toxic effects, such as hypersensitivity reactions, thrombosis, pancreatitis / pancreatic necrosis, hepatotoxicity. For safety profile improvement a technology pegylation was lay down PEG-aspargase drug produced. This has less effects recommended first-line leukemia. Drug monitoring for assessment the effectiveness toxicity optimal. Such individualization helps dose finding decrease frequency severity effects.

Transient hyperthyroidism following L-asparaginase therapy for acute lymphoblastic leukemia.
The effect of L-asparaginase on the thyroid gland has not been well documented. We report the first two cases of hyperthyroidism associated with thyroid nodule following L-asparaginase therapy for acute lymphoblastic leukemia (ALL). The thyroid function abnormalities were not severe, short-lived and did not require specific therapy.

متن کاملHypoglycemia associated with L-asparaginase in acute lymphoblastic leukemia treatment: a case report
A patient with acute lymphoblastic leukemia repeatedly developed hypoglycemia during chemotherapy. Comparison of serum glucose trends between chemotherapy with and without L-asparaginase (L-Asp) demonstrated a strong association between L-Asp and hypoglycemia. Critical blood sampling during hypoglycemia indicated hyperinsulinism, suggesting that L-Asp induced hypoglycemia in the patient through...
متن کاملUse of L-asparaginase in acute lymphoblastic leukemia: recommendations of the Polish Adult Leukemia Group.
L-asparaginase is a hydrolase that catalyzes the conversion of L-asparagine--an endogenous amino acid necessary for the function of some neoplastic cells, such as lymphoblasts. In most human cells deficiency of L-asparagine can be compensated by alternative synthesis pathway through which L-asparagine is produced from aspartic acid and glutamine by asparagine synthethase. Depletion of L-asparag...
